What does CGN stand for?
CGN stands for Common Gα Numbering, the Gα protein numbering system described in Flock et al., Nature (2015).
What do the letters and numbers of the CGN stand for?
The CGN positions are given in the following format: D.S.P – D describes the domain (G for GTPase domain, H for Helical Domain) the position is located in, S stands for the secondary structure element the residue is located in and P is pth position within the consensus secondary structure element.
How should I use the CGN for my G protein of interest?
We recommend using the CGN as a superscript for individual positions of specific G protein. For instance, position Phe336 in Gαi can be represented as Phe336H5.8, which is the 8th position from the start of the consensus Helix 5 in the human reference (Domain optional: Phe336G.H5.8). The corresponding position in Gαs would be Phe376H5.8. If no particular Gα protein is referred to, PheH5.8 is sufficient to describe the position.
How many sequences and structures does the CGN include so far?
The current database covers around 1000 Gα protein sequences from different species and 80 different Gα structures.
I cannot find my sequence in the CGN database, what shall I do?
You can upload your own sequence(s) in fasta format. Up to 20 sequences can be submitted and mapped to the CGN at a time.
